Intracellular homologous recombination is a commonly used method to insert the gene of interest into baculovirus DNA in BEVS. However, the recombinant rate can be unstable sometimes. BaculoGold expression system applies the so called “rescue strategy” to solve the problem. The viral DNA is engineered with a lethal deletion. Only when the BaculoGold DNA is co-transfected into the insect cell and homologous recombinant with the transfer plasmid which contain the segment of target gene, can the recombinant virus DNA survive in host insect cell.

The basic BaculoGold Expression kit (KTE1011) contains the modified linearized BaculoGold Baculovirus DNA, transfer vector set as well as other essential components. Except that, Creative BioMart also provides "all-in-one" 6xHis and GST Expression and Purification Kit (KTE1012, KTE1013) for tagged protein expression and purification.
In addition, the recombinant and infection process typically happen inside the insect cells which is invisible and cannot be monitored. Creative BioMart provides a Bright Linearized BaculoGold system in which the baculovirus DNA framework is fused with a GFP tag. By fluorescence microscopy or flow cytometry, the recombinant viruses can be detected, allowing the study of kinetics of infection and protein expression.

An array of versatile transfer vectors can be used as the transfer vector for the BaculoGold system. According to different requirements, we provide vector with single or multiple polyhedrin or p10 promoters for 1-4 proteins expression simultaneously. These vectors can also be used to express protein with singly or multiply 6xHis or GST tag or secretory protein with signal peptide. Additionally, vectors are available for easy visualization of GFP-, BFP-, or YFP-tagged proteins.
