Protein is the basic material of life and the basic organic matter of cells. It should be noted that most proteins do not perform their functions as monomers. In most cases, multiple protein subunits’ function can be switched on only when they are combined with some peptides to form protein complexes. However, these complexes are not always readily available in natural condition. Some complexes exist only in trace amounts in cells, while others degrade rapidly after formation.

In order to facilitate the study of protein function and structure, it is a good choice to obtain these complexes by eukaryotic expression system. Baculovirus expression system has become a powerful expression tool because of its large foreign gene carrying capacity and post-translational modification ability. However, the insertion of multiple genes makes the construction of expression vector a problem. The traditional vector construction process relying on multiple cloning sites (MCS) has the short comes such as low flexibility, slow speed and difficult screening. The above factors not only restrict the expression of protein complexes as well as the protein research field.

Technology Description

MultiBac Expression Approach

• Description: An innovation system which is based on the conventional Bac to Bac expression.
• Feature: Adopt homing endonuclease /BstXI cloning + Cre-Lox recombination to complete multi gene insertion rather than conventional MCS. Use Tn7 transposition to construct expression vector inside the E. coli strain.
• Applications: Can mediate the insertion of foreign genes cassettes to vectors efficiently and flexibly. The number of inserted gene is theoretically unlimited.

biGBac Expression Approach

• Description: A combination system of baculovirus Bac to Bac expression and Gibson assembly reaction technology. 
• Feature: Construct only one ultimate vector via the Gibson assembly. The recombinant protein complexes exhibit improved yield, homogeneity, reproducibility and biological activity.
• Application: Suitable for the rapid flexible insertion and production of up to 25 gene subunits.

BacMam + multi gene expression approach

• Description: A hybrid system which facilitates multi heterologous DNAs delivery to mammalian cell using baculovirus.
• Feature: Similar to MultiBac Expression Approach. but the ultimate host cell line is mammalian cell line. Adopt homing endonuclease /BstXI cloning + Cre-Lox recombination to complete multi gene insertion rather than conventional MCS. A mammalian promoter is introduced into the vector.
• Applications: Can mediate the highly efficient multi gene expression in a wide range of mammalian cells.
