Glycosylation is a form of post-translational modification. It is an important modification of protein, which can regulate protein function. The importance of N-linked glycosylation is becoming increasingly obvious in pharmaceutical field. Many therapeutic proteins on the market are antibodies, N-linked glycoproteins.
Baculovirus expression system (BEVS) has gradually become one of the four eukaryotic expression systems after 30 years of rapid development. Except for the characteristics of large capacity, simple culture conditions, and high yield, BEVS is comparable to the advanced eukaryotic expression system in terms of its post-translational modification ability. The activity of protein expressed by BEVS is usually similar to that of natural protein.
However, the BEVS has limited N-linked glycosylation ability which restrict the production of some therapeutic mammalian protein via insect cell.
In order to solve this problem, Creative BioMart has established several expression pathways that can complete N-linked glycosylation, providing expression services for customers in need. According to the property of target proteins, services can be divided into two types,
Choosing our N-linked Glycoprotein Expression Service, customers only need to provide the coding sequence of your target protein. We will provide customizes service from codon optimization to large-scale expression and purification. For more assistance, please contact us for help.
Three cellular N-linked glycoprotein expression services with different strategies are offered by Creative BioMart. Each has its unique characteristics as well as drawbacks. Customers could click the links below to have a look at the working principle and service detail of each strategy. If you need any assistance on pathway selection, please contact us for help.
Co-infection the host insect cells with the recombinant viruses harbour target gene and glycozymes sequence
Integrate the stable open reading frames (ORF) of glycozyme into the genome of a host cell.
For secertory N-linked glycoprotein expression, sf9 cell line is not appropriate any more. Cell lines derived from Trichoplusia ni is reported to show a significantly higher protein yield in this case.
In the Secretory N-Linked Glycoprotein Expression approach, a glyco-module is introduced into the baculovirus genome, enabling the correct mammalian N-linked glycosylation of the target protein. This expression pathway based on the multi gene expression system uses a very flexible recombination method to facilitate gene insertion. It is a flexible tool for glycoengineering.
