Kinases is an enzyme that catalyzes phosphorylation process for specific substrates such as proteins, lipids, carbohydrates, etc. Phosphorylation plays an important role in metabolism, cell signal transduction, protein regulation, transportation and secretion. Many diseases and even cancer are associated with kinase dysfunction.
To facilitate the research of kinase related cancers and kinase regulatory drugs, high-quality recombinant kinases need to be provided as an important target protein. In the pharmaceutical and biotechnology industries, protein kinases are mainly produced by baculovirus expression systems, considering the factors of applicability, cost and yield. A package containing the ORF of 7 kinase chaperones was inserted into AcMNPV gene via the loxP site. These protein chaperones contribute to the folding of kinase enzymes, thus enhancing the stability and yield of functional proteins. Through the manipulated kinase expression approach, we can produce those previously unobtainable kinases for our customers at high levels, in soluble and functional form.

The expression of KRS1 was detected by SDS-PAGE with Coomassie staining. The left lane is the unmodified, and the one on the right is the modified kinase expression system. T is the total cell extract and S is the soluble protein. As shown in the figure 2, the kinase is almost invisible in the expression pathway without chaperone (the lift two). On the contrary, a high level of kinase expression was observed in the modified system.
