Even a small virus provides a huge surface, which can be manipulated in many ways. Baculovirus surface display (BSD) system one of the novel eukaryotic surface display systems developed in recent years. The target protein or polypeptide can be displayed on the surface of baculovirus by fusing with glycoprotein gp64 on baculovirus surface or binding with specific anchoring site. Due to the feature that foreign protein still maintains its original activity after the display, BSD system have become valuable tools in biotechnology and medicine.
In the BSD system, the foreign proteins are typically displayed either on the surface or the infected cell surface through fusion with viral glycoprotein GP64 (A). Generally, there are several location options for the foreign protein to be fused with—— for example the GP 64 siginal peptide (SP, B), GP 64’s transmembrane (TMD, C), GP64’s cytoplasmic tail (C-terminal domain, CTD, D), or both the TMD and CTD domains of GP64 (E).
In addation to GP64, the VP39, a major fusion partner of baculovirus’ capsid protein can also be used to conduct surface display.Based on the infusion partners (GP64 and VP39), the forgien protein can be displayed on the surface of baculovirus, baculovirus capsid, baculovirus occlusion body, infected insect cell and mammalian or other non-lepidopteran cell.
