Baculovirus is a kind of virus which takes arthropod as a single host. Its genome has a large capacity and can accommodate the insertion of 38 kb foreign gene fragments. Studies have shown that wild baculovirus can enter mammalian cells but will not turn on the expression of its own protein, unless the mammalian promoter is inserted upstream of that gene. However, baculovirus can not replicate in mammalian cells whether it has been inserted into a recognizable promoter in mammalian cells, so it is non infectious. This feature of the baculovirus was made into a novel technology in the 1990s, which uses baculovirus to transfer foreign genes to mammals, as an improvement of the traditional baculovirus expression system, named BacMam.
Ability of baculovirus to enter mammalian cells means that transduction can be achieved by simply adding viral inoculums into cells, which have been shown to be highly efficient and dose-dependent. Moreover, the BacMam baculovirus can efficiently transduces most mammalian cells – including primary cultures, cell lines, and iPSC derived cells. Therefore, since its development, BacMam has been widely used in various fields of biotechnology and medical treatment.
