Ultra-Stable Baculovirus System for Expression Scale Up- SynBacMultiBac is one of the milestone expression pathways in baculovirus expression system. The establishment of this multigene expression strategy has facilitated the expression of protein complex, extending the application of BEVS to more fields (like vaccine development and protein structure study, etc.). However, a disadvantage of MultiBac is that the virus genome is unstable during amplification. After several generations, the foreign fragments will be gradually eliminated, which seriously affects the expression of heterologous proteins. For academic studies that usually require only a few milligrams of protein samples, this problem can be alleviated by avoiding virus amplification.

However, considering other application fields of MultiBac that require scale-up (vaccine production, etc.), a redux MultiBac expression approach——SynBac is applied to solve the problem of virus DNA eliminating. 

TECHNOLOGY DESCRIPTION

SynBac is similar to that of multi gene expression approach. The two approaches share the same expression principle, vectors as well as host cells. In general, SynBac is a redux approach of MultiBac, which support stable large-scale protein expression. The only different between them is the component and size of virus genome. According to the genome analysis, many gene cassettes in wild AcMNPV genome are invalid or harmful to the expression of target protein. Researchers segmented the genome and deleted those fragments. After many times of connection and expression testing, a simplest genome of baculovirus was synthesized. Experiment highlights the excellent performance of SynBac after subculture.
