After the virus stock amplification, the recombinant baculovirus can be used to infect insect cell lines to activate recombinant protein expression. However, even for the mature expression pathway which has witnessed successful protein expression for many times, small-scale protein pre expression is still essential before scale-up.

WHY CONDUCT SMALL-SCALE EXPRESSION AND EVALUATION

• Baculovirus expression is an invisible process.

Undesired events such as abnormal coding sequence mutation of target protein and vector construction failure are often unpredictable. In this case, it would be imprudent to directly carry out large-scale protein expression. In order to avoid the loss caused by these undesired factors, small-scale recombinant protein expression is usually carried out first.

• The recombinant protein was sequenced and identified to confirm that the current expression system is well-established, and the target protein could be produced normally.
• The expression capacity of the expression approach will be tested in this step, which can provide the basis for the subsequent system optimization.
• The small-scale expression and evaluation step will be the final step for customers who only need few amounts of target protein.
