The acidic glycoprotein gp67 is a major envelope protein of baculovirus particles and plays an essential role in virus entry into insect cells. Its signal peptide is one of the most efficient secretion signals encoded by baculoviruses. Based on this property, baculovirus transfer vectors have been constructed in which the gp67 signal sequence is positioned upstream of a multiple cloning site. Insertion of a gene of interest into this site enables expression of the recombinant protein as a gp67 signal peptide fusion, promoting efficient secretion into the culture medium under the control of a strong baculovirus promoter. This strategy facilitates recovery and purification of secreted recombinant proteins from insect cell cultures grown under serum-free conditions. |
