pFastBac1-C-EGFP is an insect cell expression plasmid designed for C-terminal fusion of EGFP to a target protein. Expression is controlled by the baculovirus polyhedrin promoter, allowing high-level production of the target protein–EGFP fusion in insect cells such as Sf9 and Sf21. EGFP contains dual affinity tags, enabling purification by metal-affinity or alternative ligand-based chromatography. A site-specific protease cleavage sequence is positioned between the target protein and EGFP, allowing removal of the fusion partner and affinity tags after purification. When seamless cloning strategies are applied, protease cleavage leaves only minimal residual residues at the protein terminus. The EGFP fusion enables real-time monitoring of protein expression during viral infection, facilitating optimization of harvest timing. The dual-tag configuration provides flexible purification options under mild conditions while maintaining protein integrity and activity. This plasmid is compatible with commonly used baculovirus-mediated insect cell expression workflows. |
