pFastBac1-EGFP is an insect cell expression plasmid engineered for the expression of enhanced green fluorescent protein (EGFP). Expression is driven by the strong baculovirus polyhedrin promoter, enabling efficient EGFP production in commonly used insect cell lines such as Sf9 and Sf21. When applied in a transposition-based baculovirus expression workflow, this plasmid can be used as a positive control to monitor plasmid transfection, recombinant virus generation, and target protein expression. Robust green fluorescence allows convenient visualization of these processes by fluorescence microscopy. The vector functions as a transfer plasmid in which the gene of interest is cloned downstream of a baculovirus-specific promoter. Introduction of the recombinant plasmid into a suitable E. coli host carrying a bacmid and transposition machinery enables site-specific insertion of the expression cassette into the bacmid genome. Recombinant bacmids can be identified by disruption of a reporter gene, confirmed by PCR, isolated, and subsequently transfected into insect cells for recombinant protein expression. |
