pFastBac1-N-EGFP is an insect cell expression plasmid designed for N-terminal fusion of EGFP to the target protein. Under the control of the baculovirus polyhedrin promoter, the EGFP–target protein fusion is efficiently expressed in insect cell systems such as Sf9 and Sf21. Strong EGFP fluorescence enables direct visualization of protein expression by fluorescence microscopy. The EGFP moiety carries dual affinity tags to facilitate purification using metal-affinity or ligand-based chromatographic methods. A protease cleavage site located between EGFP and the target protein allows removal of the fusion tag after purification, leaving only minimal additional residues when optimally cloned. N-terminal EGFP fusion supports real-time assessment of expression levels during viral infection, aiding optimization of virus amplification and protein yield. This plasmid is suitable for use in standard baculovirus-based insect cell expression workflows. |
