Technology Description

The Bac-to-Bac HBM TOPO secreted expression system is an innovation system based on the Bac to Bac TOPO expression system. Similarly, it consists of

  • A donor vector
  • Contains a N-terminal Honey Bee Mellitin (HBM) secretion signal coding sequence to facilitate the recombinant protein secretion into the extracellular medium.
  • Also contains a C-terminal polyhistidine tag that allows easy purification of the secreted protein.
  • E. coli host strain which contains a bacmid.

The gene fragment of interest can be cloned to donor vector via the blunt-end TOPO cloning technology in only 5 mins. The gene fragment is then inserted to bacmid from donor vector through Tn7 site-specific transposition. After bacmid infecting insect cell lines, the recombinant protein could be collected, directly in the culture medium.
