Almost all baculovirus expression pathways involve the step of integration of plasmid vector and virus DNA. The " integration " here mainly refers to Tn7 transposition recombination occurred outside the host insect cells and homologous recombination occurred inside the host insect cells. Of course, Creative BioMart's expression platform also provides some other expression pathways using other recombination methods (such as LR recombination, etc.).

Each integration method has its own suitable transfection strategy. Creative BioMart provides customized transfection services which are corresponding to different integration methods. After the virus generation, customers can choose to authorize us to perform subsequent protein expression services. If you choose to terminate the service, we will deliver the virus stock to you in the first time. Please know that whenever you need help, Creative BioMart will always be there.

Tn7 TRANSPOSITION RECOMBINANT-BACMID TRANSFECTION SERVICE

For customers who want to use transposition recombination to form novel baculovirus, Creative BioMart offers bacmid transfection service. Baculovirus DNA and donor vector were recombined into the recombinant bacmid in host E. coli. After bacmid extraction, insect cells were transfected with transfection reagent.

General process

• Recombinant bacmid is extracted from the E. coli host cell
• Bacmid is transfected into the insect host cell
• Positive virus screening

Note. Although BaculoDirect expression pathway does not construct vector by transposon recombination, it is still integrated outside insect cells, so it can be introduced into insect cells by ordinary transfection program.

Application

HOMOLOGOUS RECOMBINANT-CO-TRANSFECTION SERVICE

Co-transfection is an essential step of recombinant baculovirus construction. Creative BioMart offers a cost-effective approach to customers who wish to ensure success with their baculovirus co-transfection.

General process

• Provide a transfer plasmid containing the target gene

Provided by the customer or produced by Creative BioMart vector gallery.

• Co-transfection

Transferred plasmid is then used with different forms of baculoviral DNA (circular or linear) to co-transfect insect cells.

• Control co-transfection

For those expression pathways which use blue and white spot screening (the transfer plasmid contains the lacZ gene), a control co-transfection will be conducted.

• Send virus stock (P0) to the customer.
