CODONS OPTIMIZATION

Codon optimization refers to the redesign of genes by using preferred codons which avoiding rare codons with low utilization, simplifying the secondary structure of mRNA transcription, removing motifs that are not conducive to efficient expression, adding motifs that are conducive to efficient expression, and adjusting GC content. For the codon optimization, we mainaly focus on the following aspects

  • Codon Usage Bias
  • Adjust rare codon components. Combining high-frequency and sub high-frequency codons to make the codon adaptation index (Cai) reach Cai ≥ 0.80 (which is the theoretical high-efficiency protein expression standard).

  • mRNA Secondary Structure
  • Identify and reduce hairpin structure.

  • Removal of Trans Acting Elements and Restriction Sites Affecting Expression
  • Selective deletion of restriction sites in order to avoid the impact of site conflict on the construction of expression vector.

  • GC Content
  • GC content directly affects the binding stability and annealing temperature of DNA sequences

  • Effective translation termination can promote protein expression.
