Baculovirus can not replicate in mammalian cells and does not cause cytopathy. It is a reassuring tool for exogenous gene delivery. Double-stranded baculovirus DNA can effectively transfers and expresses the gene of interest in mammalian cells. BacMam particles (recombinant virus) are absorbed by cells through endocytosis and migrate to the nucleus to release genetic material for transcription and expression. The expression of exogenous genes can be detected from 4-6 hours to 5-14 days after transduction, depending on cell type, division rate, and transduction efficiency.
Tn7 mediated (Bac to Bac) BacMam vector construction is best-developed method but the process of site-specific transposition can be time-consuming. To improve the expression efficiency, BacMam + Gateway Expression Approach is now avaliable in Creative BioMart.
Gateway technology can facilitate the insertion of the gene of interest into the baculovirus genome in vitro, eliminating the section of transforming bacteria, bacmid extraction, or co-transfection. Through the Gateway recombination reaction, the recombinant baculovirus genome needed for transfection can be obtained within one hour, dramatically shortens the protein expression period.
