Protein Quantitative Analysis Service

Protein quantitation is often necessary before processing protein samples for isolation, separation and analysis by chromatographic, electrophoretic and immunochemical techniques. Depending on the accuracy required and the amount and purity of the protein available, different methods are appropriate for determining protein concentration.

OPTIONS FOR QUANTITATIVE ANALYSIS SERVICE

Here are some quantitation methods available at Creative BioMart. But please note that no method is perfect for every protein. The advantages and disadvantages are listed below. Customers can choose according to the instructions or contact us for help.

UV absorption

Simplest and most direct assay method for protein concentration determination

  • Simple, no reagents needed
  • Highly error prone with protein mixtures or complex samples (e.g. cell lysates)

Biuret methods:

Protein-copper chelation and secondary detection of reduced copper

  • Compatibility with most surfactants (detergents)
  • Good performance of linear response curve (R2 > 0.95)
  • Less protein–protein variation than the Coomassie dye–based assays
  • Incompatibility with substances that reduce copper or common reducing agents such as DTT

Colorimetric dye-based methods: 

Protein-dye binding and direct detection of the color change

  • Fast and user-friendly
  • Performed at room temperature
  • Compatible with most salts, solvents, buffers, thiols, reducing substances, and metal-chelating agents
  • Incompatibility with surfactants (detergents)
  • High protein–protein variation when compared to copper-based assays

Fluorescent dye methods: 

Protein-dye binding and direct detection of increase in fluorescence associated with the bound dye

  • Excellent sensitivity, less protein sample required for quantitation
  • Assays can be adapted for automated handling in high-throughput applications due to less significance of timing

  • Specialized equipment required
